AAV vectors: Basics and Current Applications in Gene Therapy

企画・趣旨のねらい 山形崇倫（Takanori Yamagata） 自治医科大学小児科 小坂　仁（Hitoshi Osaka） 自治医科大学小児科 　中枢神経疾患に対する遺伝子治療開発が世界的に，急速に発展し，臨床応用も開始されている．中心は，アデノ随伴ウィルスベクター（AAV），レンチウィルスベクター，small RNAである．日本で開発が進められている遺伝子治療の現状報告と，将来の展望について議論を深め，他の疾患も含めた遺伝子治療開発における共同研究，臨床応用時の連携促進を図りたい． 　村松慎一氏にAAVの基礎知識と世界的な状況について説明していただき，応用可能な疾患を模索したい．小坂　仁氏には，AADC欠損症に対する遺伝子治療の臨床研究経過の報告と，GLUT1欠損症に対する治療法開発の現状を報告いただく．伊藤孝司氏にTay—Sachs病に対するAAVを用いた遺伝子治療開発の現状と臨床応用の展望を，井上　健氏にPelizaeus—Merzbacher病に対する人工miRNAを搭載したAAVによる遺伝子発現抑制治療開発研究について，新堀洋介氏にDravet病のモデルマウスの解析とそれを踏まえた遺伝子治療法開発研究について報告いただく． 　いずれも，難治性疾患の治療として期待が大きい研究であり，他の難治性疾患の治療法開発に道を開くものである．
