Escherichia coli (E.coli) is an index for hygiene and pollution of water. We are developing an optical disk device that can detect the presence of a single E.coli without its cultivation. Since we found by numerical calculations that transmitted light gives a clearer and more distinct difference between E.coli and other objects than reflected light, we have improved the device so that we can measure the transmitted light intensity, and have succeeded in detecting a single E.coli.
