In the current study, we used guanine, inosine, and cytosine as the counter bases and CNVK, OHVK, and NH2VK as the cross-linker. As per our hypothesis, the increased hydrophilicity increased the reaction rate and the pair of inosine as counter base and OHVK as the cross-linker showed the highest rate of reaction with a half-life of 4 days. We further carried out the mutation of short oligo-RNA and was able to achieve the similar results. Thus, this technique can be used for potential in vivo editing of both DNA and RNA.
