[P88]Synthesis of cyclic oligo (ethylene glycol) for suppression of amyloid insulin fibrillation

Insulin fibrillation is a major problem in type 2 diabetes. Therefore, we attempted to synthesize cyclic amphiphilic OEG molecules oligo (ethylene glycol) to suppress insulin fibrillation. Each step of molecule confirmed by NMR and MALDI-TOF mass spectrometry. The insulin solution was incubated with cyclic OEG at 60 oC in phosphate buffer pH 2 for 8 hours. But insulin in the presence of monomer with weight ratio 1:16 and 1:10, 80% of healthy insulin was remained after 8 hours. Thus, this study demonstrates the importance of amphiphilicity in the design of cyclic OEG to suppress insulin fibrillation.
