我々は先行研究で、普遍遺伝暗号と直交性を持つSer/Leu交換遺伝暗号を開発した。本系で用いるSer/Leu交換遺伝子は、通常の生物では機能性タンパク質に翻訳されず、バイオハザードのリスクが軽減される。一方、本系はタンパク質合成量が少ないという問題があった。そこで本研究では、天然・転写tRNAを用い、Ser/Leu交換遺伝暗号を持つ高活性な無細胞翻訳系の構築を行なった。
