Engineering principles applied to harness biology incite technologies accessing goods and products of properties and quantities limited only by imagination. The extent to which the principles themselves can be harnessed are limited by the reproducibility of their application to biological systems. An ultimate test to link engineering and biology is that of our ability to code biology into chemical space, beyond the perceived constraints of classical protein engineering and the central dogma. Here engineering approaches to expand biology with biologically orthogonal chemical topologies to design de novo functional assemblies are discussed. Examples include virus- or phage-like systems, and other systems designed to elicit specialist biological functions, similar and atypical of naturally occurring analogues.
