VTT's Biofoundry accelerates industrial biotechnology research through modular strain generation and laboratory automation for high-throughput engineering and characterization of bacterial and fungal strains. We routinely work with species such as Trichoderma reesei, Pichia kudriavzevii, Saccharomyces cerevisiae and Escherichia coli. Our in-house developed synthetic biology toolkits, modelling pipelines and AI-tools allows us to design and engineer new enzymes and pathways for production of e.g. proteins, bioplastics (PHA), pigments, lipids, chemicals or materials. Here, the design-build-test-learn (DBTL) cycle for creation of cell factories for material applications will be presented. The combination of modelling and active learning during the DBTL cycle allows us to increase our success rate. After the primary strain engineering and screening phase, the best strains move towards process development in controlled bioreactors where process analytical technology (PAT) such as Raman spectroscopy, on-line microscopy, refractometers, EMF sensors and FTIR analysis can be used to better understand the cultivation process.
