Our group interests are in the broad areas of enzyme catalysis, enzyme and metabolic engineering. As fundamental understanding of enzymatic reactions can effectively support engineering methodology to improve properties of biocatalysts or engineered cells, we often combine mechanistic enzymology and enzyme engineering via semi-rational or mechanism-guided approach to engineer enzymes which has resulted in successful improvement of biocatalyst properties with efficient resource utilization. In this talk, I will discuss our recent work in using ancestral sequence reconstruction to create decarboxylases which can be used in chemo-enzymatic C-C bond coupling reactions. We also used mechanism-guided enzyme engineering and machine learning approach to engineer a phenolic hydroxylase to be enantio-selective and demonstrated use of the engineered enzyme for synthesis of catecholic drugs.
