CRISPR-based technologies have revolutionized biomedical sciences and biotechnology by enabling researchers to modulate the genome of living cells at will. However, existing tools suffer from several shortcomings that remain to be addressed. For example, while base editing provides a means to install single nucleotide changes in the genome readily, it can generate excessive RNA off-targets and may also pose other safety issues. Additionally, while CRISPR-mediated homology-directed repair (HDR) enables the integration of large DNA payloads, it remains inefficient in mammalian cells. Here, I will describe our efforts to characterize genome engineering tools more carefully and will present some strategies to address underlying problems.
