[Purpose] Developing new genome editing methods for archaea and bacteria

[Method] CRISPR-based/TnpB-based gene-editing tools and RedET genetic engineering

[Results] Our research on RNA-guided TnpB nucleases has enabled gene-editing at single nucleotide level both in archaea and in bacteria. In addition, we have also developed a new genetic scheme for identification of unknown essential and quasi-essential genes in microbial genomes and replace the original genomic segment using synthetic DNA fragments carrying assembly of essential genes. We also demonstrate that utilizing both Cas9 gene-editing and RedET recombineering further facilitates the genome-trimming process in bacteria.

[Consideration]

[Conclusion] New gene-editing methods have been developed for both archaea and bacteria and they can be tested in other archaea and bacteria.
