All of developed microbial cells have to cultivate in production media, which compose of various chemicals including carbohydrates, phosphate, nitrogen, sulfate, minerals; additionally, amino acids, nucleotides, and vitamins in some cases. Natural feedstocks including molasses, corn steep liquor, meet extract, yeast extract, peptone etc. are often used to satisfy the nutrient requirement of the cells due to the low cost and easy handling. However, the detail compositions of the natural feedstocks have been unclear. Therefore, the differences among the production lots and/or brands often cause troubles in the cultivation processes such as low growth and productivity. In our study, to solve the problems in uses of natural feedstocks, the correlation analysis between performances and compositions of the natural feedstocks were performed by multimodal targeting analyses with an amino acid analyzer including free and total amino acids, ion chromatograph, liquid chromatography-mass spectrometry, and inductively coupled plasma mass spectrometry. The number of the targeted compounds was 102, including salts, minerals, amino acids (peptidic and free amino acids), nucleic acids, organic acids, and vitamins. To investigate the profiling performance of the analyses, 36 different peptones were analyzed. Seventy-eight compounds/ions were detected the peptones and total sum of the quantified materials were exceed 65% of total weight in all cases. Based on the principal component analysis, plant-derived peptones showed the greater varieties in the composition than animal- and casein-derived peptones. In the view of the difference of processing, acid-digested peptone included free amino acid greater than enzyme-digested ones. In the next, significant components in peptone and yeast extract were investigated for sophorolipids (SL) production by a yeast Starmerella bombicola. The correlation analysis, using partial least square regression, between SL-productivity and the composition in the peptone and yeast extract revealed the significant components to improve the SL production. The adducts of amino acids cocktails, which proposed by the statistical models truly improved SL production in a poor performance production medium. We believe that the methodology can be applied to versatile of feedstocks and bioprocesses.
