[Background] Indonesia is among the 17 countries worldwide that are considered to be megadiverse1). Nonetheless, the substantial microbial diversity present in these regions has not yet been fully tapped for drug discovery, primarily due to limitations in local research infrastructure. In collaboration with BRIN (Indonesia's National Research and Innovation Agency) and Hasanuddin University, actinomycete and fungal strains were screened for the discovery of novel bioactive compounds.

[Method] In this study, actinomycete strains (IDNA) were cultivated in two M1 production media, with or without deep-seawater. The selection of the strain was made on the basis of an analysis of the secondary metabolite chromatography profiles. The cell cytotoxicity was evaluated using MTT colorimetric method.

[Results and Discussion] Strain IDNA-0041 was selected and cultivated in M1 medium with seawater, which resulted in a significant increase in the production of secondary metabolites. The 7-day culture broth was filtered and the supernatant was refined using HP-20 and then ODS column. The 40% MeCN ODS fraction was subsequently subjected to preparative HPLC, yielding two novel compounds (1 and 2). Structures of 1-2 were determined through a series of spectroscopic experiments, including MS and 1D/2D NMR. Both are bridged angucyclines, with 1 featuring a rare amino sugar moiety. The structures are considered particularly intriguing from a biosynthetic perspective. Furthermore, these compounds showed cytotoxic activities against HeLa and HepG2 cells.
