[Purpose]
Efforts to discover novel bioactive natural products from Streptomyces biosynthetic gene clusters (BGCs) have gained increasing attention. However, specialized metabolite production based using BGCs remains labor-intensive and time-consuming due to the inefficiency of conventional experimental methods in their native hosts. Hence, advanced synthetic biology tools and workflows are introduced to streamline and accelerate genetic manipulation.
[Method]
An optimized CRISPR–Cas9BD system for Streptomyces was developed by adding aspartate residues in both sides of the protein.
[Results]
The newly engineered Cas variant exhibited reduced cytotoxicity across various actinomycetes, thereby simplifying BGC genetic modification. Using this system, diverse workflows for genome editing, such as refactoring and deletion, were successfully developed.
[Conclusion]
This system improved the utilization of Actinomycetes as a host for producting specialized products.
