The development of technologies for the rapid and highly accurate detection of target compounds is critically important in the Test phase of the “Design–Build–Test–Learn” cycle in synthetic biology and metabolic engineering. We are developing a novel biosensor in the form of a chimeric protein by genetically fusing fluorescent proteins with proteins that have high affinity for target compounds. This biosensor enables the detection of conformational changes induced by the presence or absence of the target compound through fluorescence output. This system has broad applications, including the quantitative analysis of valuable compounds produced by microorganisms and enzymatic reaction products, and therefore contributes to accelerating the Test phase.
In this study, we reports novel biosensors for malonyl-CoA, a key metabolite in fatty acid and polyketide biosynthesis, and cis,cis-muconate, an important chemical intermediate in the production of plastics and other chemicals. Using these biosensors, we established new methods for measuring the activity of enzyme sets responsible for the production of malonyl-CoA and cis,cis-muconate.
