[P01-084]Characterization and metabolite analysis of the Streptomyces sp. isolated from "Yukido" lava tunnel in Daikonshima

There are two lava tunnels known as Nationally Designated Natural Monuments in Daikonshima in Matsue-City, Shimane Pref., Japan. Especially, "Yukido" is designated as Special Natural Monument and preserved by the Cultural Heritage Division. At the lava rock wall of Yukido, two independent microbial communities (yellow and white) were observed. They are divided by distinct smooth boundary, indicating the presence of antimicrobial substance(s) to maintain their own territories at the lava rock wall. Hence, we performed the isolation of microorganism attached at the ceiling.
White-color attachments at Yukido were suspended in phosphate buffer, inoculated on Humic acid-Vitamin agar (HV agar) plate, and incubated at 25ºC. The growing colonies, which appeared to be Actinomycetes, were transferred onto the new HU agar plate, and Actinomycetes-like microorganisms were obtained. The partial 16S rDNA sequence analysis revealed that the isolate was member of Streptomyces. So, this isolates was named "YU-W-01" and proceeded with the analysis. To characterize this strain in detail, we performed whole-genome sequencing of the strain YU-W-01. The genome sequencing indicated that the genome of this strain consists of one linear chromosome, which harbors about 8.64 Mb nucleotides with 72.1 % GC content, together with three contigs, possibly plasmids. From the result of pairwise comparison analysis, the strain YU-W-01 was closely related to Streptomyces nojiriensis, with around 90 % of dDDH value.
The result confirmed secondary metabolite production showed that strain YU-W-01 produces anti-microbial material against Bacillus subtilis and Micrococcus luteus. Moreover, antiSMASH analysis revealed the presence of 21 biosynthetic gene clusters (BCGs) on the chromosome of strain YU-W-01. Further works, including structural analysis of antimicrobial compounds and genome structure, are in progress.
