The escalating threat of antimicrobial resistance and the persistence of tropical diseases like dengue and malaria necessitate the urgent discovery of novel therapeutic agents. This research, a multilateral collaboration between the Philippines, Vietnam, and Indonesia, targets the untapped chemical diversity of marine-derived Actinomycetes. This research aims to isolate and perform structural elucidation of bioactive compounds produced by Actinomycetes from mangroves and coral reefs. A total of 160 Actinomycete strains were collected and results showed that 114 of the 160 isolates exhibited considerable bioactivity, with zones of inhibition (ZOI) ranging from 10 to 40 mm against pathogenic bacteria. Subsequent analysis of their Thin Layer Chromatography (TLC) profiles was performed. Interestingly, TLC bioautography of Streptomyces strain 34S-CP revealed three distinct spots with antimicrobial activity, which were further investigated. Notably, electrospray ionization mass spectrometry (ESI-MS) spectrum of strain 34S-CP showed two distinct peaks at m/z 1133.6257 [M+Na]+ and m/z 1147.6415 [M+Na]+. These molecular ion peaks showing m/z values greater than 1,000 indicate the presence of medium-size peptides, likely belonging to the cyclic depsipeptide family. Hence, its whole genome was analyzed using next generation sequencing (NGS). Results have shown that strain 34S-CP revealed the presence of 34 biosynthetic gene clusters (BGC) and interestingly, depsipeptide BGC was also detected.
