[P02-184]Solvent-Dependent Extraction and Characterization of Chlorophyll and Carotenoid Pigments from Ulva reticulata Using Spectrophotometry and LC-MS

[Purpose]
Marine macroalgae are promising sources of natural pigments with applications in food, nutraceutical, and biotechnological industries. Ulva reticulata contains chlorophyll and carotenoid pigments with potential as natural colorants and bioactive compounds. This study aimed to evaluate the effect of different solvents on pigment extraction and to characterize carotenoid profiles using an integrated analytical approach.
[Method]
Pigment extraction was carried out using ethanol, n-hexane, and acetone solvents, followed by spectrophotometric analysis.
[Results]
The results showed that pigment content varied depending on the solvent system. The ethanol extract exhibited the highest pigment yield, with total chlorophyll and carotenoid contents of 16.32 mg/g and 0.27 mg/g, respectively, followed by n-hexane with 15.66 mg/g and 0.26 mg/g. In contrast, acetone showed significantly lower pigment contents. Carotenoid profiling using liquid chromatography–mass spectrometry (LC-MS) identified 6–7 carotenoid compounds, including α-carotene, β-carotene, γ-carotene, lutein, loroxanthin, and echinenone. The chloroform extract showed the highest overall carotenoid abundance, while the methanol extract exhibited the highest relative abundance of echinenone (2.92%), indicating selective extraction of oxygenated carotenoids. The n-hexane extract showed a more selective profile with moderate abundance and absence of echinenone. These findings demonstrate that solvent type influences pigment yield and composition.
