3-Hydroxypropionic acid (3-HP) is a C3-based versatile platform chemical offering significant environmental and industrial advantages over petroleum-based derivatives; however, its commercial viability from biosynthesis is limited by low productivity and yield, primarily due to the inefficient cascade reactions of multi-enzyme systems. Here, we engineered Cip-mediated enzyme clusters to enhance 3-HP synthesis. To endow self-assembly, the enzymes involved in 3-HP synthesis were rationally engineered as fusions with CipA or CipB proteins, enabling their spatial proximity, increased local concentration of intermediates and thereby improved titer of 3-HP. Moreover, we applied this system to Vibrio sp. dhg, a marine bacterium with fast growth and a native capability to metabolize marine biomass. These findings offer a promising route toward sustainable production of 3-HP.
