Hypertension can lead to serious complications, and its prevalence has been steadily rising due to aging and dietary changes. Conventional antihypertensive drugs may cause adverse side effects, which has increased interest in bioactive peptides from natural sources. In this study, the probiotic properties and ACE inhibitory activity of Lacticaseibacillus paracasei MBE/L9125 were evaluated to assess its potential as a functional strain. rhamnosus GG KCTC5033 and L. paracasei KCTC3165, KCTC3169, KCTC3510, and KCTC13090 were used as reference strains. Antimicrobial activity was tested against Bacillus cereus, Staphylococcus aureus, Listeria monocytogenes, Escherichia coli, Micrococcus luteus, and Pseudomonas fluorescens. Acid and bile tolerance were assessed based on the growth of strains on MRS agar after 4 hours of exposure to MRS broth at pH 3.0 containing 0.3% oxgall. Hemolytic activity was determined using 5% sheep blood agar. Additionally, cells were subjected to autolysis at 55°C for 24 hours, and the supernatant was filtered through a 3 kDa membrane for ACE inhibitory activity analysis. The inhibition rate (%) was measured using an ACE Inhibitory activity assay kit (Dojindo, Japan).The results indicated that MBE/L9125 demonstrated antimicrobial activity against all tested microorganisms and produced significantly larger inhibition zones than L. paracasei KCTC3169 and KCTC3510. Furthermore, under both pH 3.0 and 0.3% oxgall conditions, MBE/L9125 showed greater growth than L. rhamnosus GG after 4 hours of treatment, with no hemolytic activity observed. In terms of ACE inhibitory activity, MBE/L9125 exhibited the highest inhibition rate among the strains tested.Overall, L. paracasei MBE/L9125 showed probiotic features and strong ACE-inhibitory activity, indicating its potential as a functional strain.
