Particulate matter (PM) is widely recognized for its detrimental impact on human health. Recently, studies have demonstrated that PM induces reactive oxygen species (ROS) generation and neuroinflammation resulting in degenerative brain disease. Fish cartilage extracts have been found to have skin protective effects. However, the effect of fish cartilage against PM-stimulated oxidative stress and neuroinflammation in BV-2 microglial cells and zebrafish have not been reported. This study aims to investigate the protective effects of fish cartilage enzymatic extracts against PM-stimulated cytotoxicity and its mechanism in BV-2 microglial cells and zebrafish. Fish cartilage was hydrolyzed with proteases to obtain enzymatic extracts. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activities of the enzymatic extracts were evaluated, and the fish cartilage Protamex extract (OCPE) exhibited the highest free radical scavenging activities. Our results showed that FCPE reduced the production of intracellular ROS, nitric oxide, and prostaglandins-E2 in BV-2 microglial cells. Furthermore, OCPE significantly suppressed the expression of pro-inflammatory cytokines and chemokines. Also, OCPE inhibited protein expression involved in triggering neuroinflammation and degenerative brain disease.In addition, OCPE potentially down-regulated PM-induced oxidative stress and inflammation by inhibiting ROS generation and lipid peroxidation in the zebrafish model. The present results suggest that FCPE could be utilized as a potential candidate in the development of pharmaceuticals by attenuating PM-induced neuroinflammation.
