Natural products play a pivotal role in the development of therapeutic drugs for human health. Recent genomic studies have revealed a vast number of untapped biosynthetic resources hidden within microorganisms. Here, we report ACTIMOT (Advanced Cas9-mediated In vivo Mobilization and Multiplication of BGCs), an enabling strategy inspired by the natural process of antibiotic-resistance gene transmission, which enables efficient mobilization, relocation, multiplication, and ultimately activation of BGCs within bacterial cells. Using ACTIMOT, we have successfully mobilized and activated 11 cryptic BGCs from Streptomyces, leading to the discovery of more than 50 compounds spanning multiple structural classes. This work highlights the potential of ACTIMOT as a powerful platform for accelerating the exploration of microbial biosynthetic pathways and the discovery of new natural products.
