Pharmacological Evaluation of Drug-Induced Liver Injury Using 3D-Cultured Human HepaSH Hepatocytes

Drug-induced liver injury (DILI) is one of the serious adverse events identified by the Ministry of Health, Labor and Welfare. During the drug development process, the risk of DILI is primarily evaluated through animal experiments in non-clinical studies; however, their predictability for humans is limited. Recent advances in the use of human-relevant tools and models, such as primary human hepatocytes, liver organoids, and humanized liver mice, have provided promising platforms for improving predictability for human DILI. In this symposium, we introduce our recent studies for evaluating DILI based on pharmacology, in addition to an overview of DILI and its toxicological mechanisms. We will also discuss the current progress and future directions in DILI research, aiming to contribute to safer drug development.
