Polarization is crucial for understanding complex optical behaviors and uncovering properties often missed by conventional methods. Traditional polarization measurement techniques usually require multiple captures, which are unsuitable for live cell imaging. This study introduces advancements in developing a polarization digital holographic microscope (PDHM) for spatially resolved, label-free imaging. Our approach emphasizes single-shot polarization imaging, greatly enhancing the feasibility of real-time observations. We demonstrate the practical application of PDHM by imaging live MCF-7 breast cancer cells, highlighting its capability to capture detailed cellular structures without the need for external staining agents.
