We investigated the structural dependency of the fluorescence enhancement in pillar-array plasmonic chip in order to improve the sensitivity in fluorescence measurements. In this study, the duty ratio and the pillar height were decreased by UV-ozone treatment, and the structural changes were measured using atomic force microscopy (AFM). To evaluate the enhancement factor in fluorescence measurements, SPR and SPF spectra of the plasmonic chips with and without UV-ozone treatment were measured and compared. The decrease in the duty ratio and the pillar height changed the resonance angle, dip depth and width in the SPR spectra, resulting in an increase in emission enhancement. These results suggested that the enhancement factor of fluorescence intensity can be improved by controlling the duty ratio and the pillar height.
