When an intracellular lipid droplet generates in an endoplasmic reticulum (ER) membrane, triacylglycerol (TAG) droplets are embedded between the phospholipid monolayers of the ER membrane. To understand this bilayer structure, we incorporated TAG droplets in giant unilamellar vesicle (GUV) bilayers. In addition to GUVs with droplets, we often observed a vesicle consisting of a single-bilayer spherical segment and a double-bilayer spherical segment. In the presentation, we discuss the reason of the formation of these vesicles.
