Because the two-photon excitation spectra of the calcium sensor and opsin are overlapping, the excitation light for photostimulation also causes fluorescence from the calcium sensor. Therefore, the background fluorescence makes it difficult to determine whether there is neural activity or not. In this presentation, we report a multiphoton imaging technique to solve this problem.
