[20a-P03-18]Surface Tension Measurements of Small Volumes of Protein Solutions

In this study, we report on the adsorption process of protein molecules at the liquid interface using quasi-elastic laser scattering (QELS). Typical surface tension measurements require 10mL to 100mL of solution, but since many biomaterials are very expensive, it is desirable to perform the experiment with as small a volume of solution as possible. Here we report on the construction of a system that can measure the surface tension of a sample as small as 1 mL by layering a low-concentration sample solution on top of a high-concentration aqueous solution. The protein used here is recombinant ferritin (N1-LF) with a carbon nanohorn-binding peptide (NHBP-1) displayed on its surface, and surface tension measurements at 37°C showed that N1-LF adsorbed on the PIPES solution and almost none on the Tris solution. These results were similar to those obtained when the experiment was performed with a large volume of solution. Therefore, the method introduced in this study was found to be effective for experiments using small amounts of solution and is expected to be applicable to the measurement of expensive or valuable samples in the future.
