Detection of the polyphenol metabolite 3-hydroxyphenylacetic acid (OPAC) was investigated using a surface-enhanced Raman scattering (SERS) substrate consisting of silver dendritic structures coated with a thin silica layer. On uncoated substrates, peaks unrelated to the analyte and broad spectral features attributed to molecular degradation and carbonization were observed under high laser power. In contrast, silica coating suppressed these unwanted signals, enabling the clear observation of multiple Raman peaks originating from OPAC.
