Although gene transfection methods using low-temperature atmospheric pressure plasma are expected to achieve both high cell viability and transfection efficiency, research on gene transfection into yeast remains extremely limited. This study aims to establish and optimize a plasma-based gene transfection method for yeast. We evaluated the effects of physical parameters in a plasma irradiation system combined with transfection auxiliary agents and heat treatment. Consequently, we found that the enhancement factor of gene transfection peaked at approximately 6.2-fold under the conditions of an irradiation distance of 5 mm and an irradiation time of 6 min.
