NEB provides NEBuilder Assembly, known as a seamless cloning method that does not depend on restriction enzymes, making it highly flexible. This method has several advantages, including the ability to clone ssDNA oligos directly and produce intact plasmids without nicks. Here, we focus on the latter benefit and present a new application workflow combining NEBuilder and its intact circular products with our phi29-XT RCA kit and NEBExpress Cell-free E. coli Protein Synthesis System. This workflow enables rapid protein expression and screening; the entire process, PCR of the target DNA to the protein expression can be completed in one day. Additionally, rapid and extensive protein screening is possible when also possible when combined with automation.
