Metal-responsive transcriptional regulations are the key features in heavy metal ion tolerance for prokaryotes.In freshwater cyanobacterium /Synechococcus/ sp. PCC 7942, /smt/ locus consists of two
divergently transcribed genes, /smtA/.( metallothionein) and /smtB/ (repressor of /smtA/transcription).
The molecular mechanism of transcription regulation and metal ion sensing with SmtB and the protein engineering techniques to improve the Zn^2+ binding ability in SmtA are studied with physicochemical techniques. We will show the results and applications of these studies.
