The application of nanoparticles (NPs) in the agricultural, medical, and industrial domains has increased and is requiring methods for monitoring plant response to NPs due to their uncertain impacts on seed germination and growth. NPs can penetrate seeds and leaves and influence cellular processes, making it crucial to understand their size-dependent effects. In this study, we proposed the Biospeckle optical coherence tomography (bOCT), developed in our lab to observe the size-dependent effect of copper oxide (CuO) NPs on the internal activity of lentil (Lens culinaris) seeds through germination exposed to CuO NPs and leaves by foliar exposure. Understanding the impact of CuO NPs is vital due to their widespread industrial use, high reactivity, potential for bioaccumulation, and significant toxicological effects on plant health and ecosystems. bOCT is a non-contact, non-destructive, in vivo monitoring technique to visualize the change of internal activity of a biological object. Our earlier study effectively confirmed its ability to assess the size-dependent effects of TiO2 and ZnO NPs on lentil seed, indicating a substantial positive impact of TiO2 NPs of 21nm and <5µm sized while ZnO NPs have a negative effect of smaller sized <50nm compared to those <5µm.
