We present a novel algorithm for measuring the optic axis of multi-layered birefringent tissue using Jones-matrix optical coherence tomography (JM-OCT). The algorithm analyzes the Stokes vector representing the polarization state of light on the Poincaré sphere to reconstruct the optic axis of the sample. The algorithm was applied to JM-OCT by converting the measured Jones matrices to the Stokes vectors corresponding to the measured polarization states reflected from the sample. We successfully validated the algorithm on single- and double-layered birefringent phantoms made of 3D printer ABS filaments, showing the possibility of applications on multiple-layered birefringent tissue.
