This paper investigates the relationship between intracellular dynamics in tumor spheroids visualized by dynamic optical coherence tomography (DOCT) and the physiological cell status determined via fluorescence (FLUO) microscopy. Our results demonstrate that the periphery of MCF-7 spheroids shows a FLUO signal, indicating “highly metabolic cytoplasm”, whereas the spheroid core shows a FLUO signal, indicating “nuclei under necrosis or late apoptosis.” Correspondingly, DOCT reveals that the spheroid periphery consists of “a high occupancy of dynamic scatterers moving at low speeds,” whereas the spheroid core consists of “a low occupancy of dynamic scatterers moving at high speeds.”
