Intracellular thermometry during freezing is important for cryobiology and cryopreservation, and NV-center fluorescent nanodiamonds provide a promising sensor platform due to their photostability, biocompatibility, and temperature sensitivity. Here, we develop a confocal thermometry platform based on NV nanodiamond optically detected magnetic resonance (ODMR) microscopy integrated with a cooling stage and demonstrate quantitative temperature measurements in frozen cells. Temperature was determined from the zero-field splitting of NV-center ODMR spectra. Intracellular 100 nm nanodiamonds maintained stable optical and spin properties after freezing, exhibiting photoluminescence count rates of 1 million counts per sec and ODMR contrast of 4–5 percent at 532 nm excitation below 1 mW. ODMR spectra recorded from 20 degC to −40 degC confirmed preservation of NV-center spin properties, establishing NV nanodiamond thermometry for thermal monitoring of frozen biological systems.
