Zebrafish are important biomedical models; however, conventional imaging techniques lack the capability for label-free, in vivo microstructural imaging. To address this limitation, we developed a custom 1.3-μm swept-source Jones-matrix optical coherence tomography (JM-OCT) system. By exploiting polarization-based contrasts, the system reveals sub-resolution microstructural features, such as pigmentation in ocular tissues, collagen-composition in swim bladder walls, the organization of muscle fibers, and pigment distribution in stripes.
