[PE2-8]1細胞発現プロファイルとバルク遺伝子発現解析の関係を表現する数理モデル

Background. Differential expression analysis between the sample groups is a standard approach in molecular biology. For example, Genes whose expression levels are statistically significantly different between disease and healthy donors are considered to be associated with that disease. Recent single-cell analysis has revealed various types of changes in gene expression profiles with cellular heterogeneity. For example, group differences can affect gene expression by working in a cellular subset-specific manner or altering the fraction of a particular cellular subset. Our research purpose is to investigate the relationship between single cell expression profile and bulk gene expression analysis.Method. We have developed a mathematical model in which the bulk gene expression value is proportional to the expected value of the probability distribution of single-cell gene expression. Based on this model, we performed theoretical and simulation analyses of the relationship between the probability distribution of single-cell gene expression and the result of bulk gene expression analysis.Result. We have identified conditions under which differences in single-cell gene expression profiles affect the differences in mean or variance of bulk gene expression. In addition, we simulated an example in which differences in single-cell gene expression profiles appear only in bulk expression variance. Conclusion. The results of this study provide useful insights for comparing the result of bulk gene expression analyses with those of single cell expression data.
