In response to the growing demand for gene transfer due to the development of regenerative medicine and gene therapy, our research group has developed a combined plasma/pulsed electric field method that combines liquid plasma discharged in a culture medium and a short pulsed electric field of 10 μs or less. By optimizing the transfection conditions, we have achieved a high gene transfer efficiency of 42.2% for blood cells, which are generally considered difficult to transfect. In this presentation, we will discuss the important factors for high transfection efficiency as well as the quantitative results of active species and electric stimulation.
