Raman spectroscopy has long been used as a powerful analytical method in the pharmaceutical field because it can non-destructively provide vibrational information of samples. For example, Raman spectroscopy can be used for the qualification of drugs in their early-stage of production, such as their crystal forms formed in different solvents, solute concentrations, and temperatures, which affect the drug efficacy; in drug development, Raman spectroscopy is used to assess drug effects and chemical changes in cells under different types and concentrations of drug treatments.
Since a large number of conditions need to be screened to find optimal conditions for producing drugs and treating cells, improving the throughput of Raman spectroscopy can extend its application. Here, we developed a multi focal spots Raman spectrophotometer for parallel observation of chemical change of pharmaceutical samples in a regular 96-well plate, which generated 96 focal spots and collected Raman spectroscopy of 96 different samples simultaneously. Using of fiber-based beam splitters and reflection-efficiency designed dichroic mirrors allows for uniform splitting of the laser beam. A lens array is used to simultaneously excite the samples and collect their Raman signals. Using multifocal spots Raman spectrophotometer, we observed crystal polymorphs, crystallization kinetics, drug response of cell spheroids, etc, showing the potential of the developed system to extend the Raman spectroscopy applications on pharmaceutical reactions.
