Raman spectroscopic imaging analyzes molecular components within a sample and provides their spatial distribution. However, due to the weak nature of Raman scattering, detecting low-concentration molecules and accurately capturing the morphology of moving samples have been challenging. In this study, we developed a cryo-Raman microscope that rapidly freezes cells and maintains a stable low-temperature environment, enabling long-term observation. This system improves the signal-to-noise ratio and resolution, and it reveals Raman scattering peaks that are not observable at room temperature. This technology is expected to enhance the utility of Raman spectroscopic imaging in life sciences and basic medical research.
