MTT, a type of tetrazolium salt, is membrane permeable and reacts with electron transporters in living cells, where it is reduced to insoluble formazan, which is deposited intracellularly. Under dark-field microscopy, more active cells produced more formazan, and the formation of red crystal particles was more pronounced. We focused on the light scattering properties of these crystalline particles and succeeded in quantifying the activity of single bacterial cells. In this presentation, we discuss the effects of various bactericidal treatments.
