In this study, we developed a single-molecule tunneling measurement technique for peptide chain terminal ends to enhance the precision of peptide sequence analysis. We explored the potential for distinguishing the N-terminal and C-terminal of peptides using dipeptide chain measurements. This technique enabled the acquisition of critical information for peptide sequence analysis at the single-molecule level. We successfully identified the terminal ends, providing a more accurate approach to peptide sequence determination. The technology has the potential to contribute significantly to the future of peptide sequencing, offering new insights into peptide structures and their functional roles in various biological processes. This method holds promise for advancing peptide research and the analysis of biomolecules, opening new possibilities for high-precision, single-molecule peptide sequencing.
