In this research, we addressed the improvement of detection sensitivity toward the higher throughput in Raman flow cytometry, which has the challenge of combining the long exposure times required for Raman spectroscopic measurements with high throughput. Specifically, we improved detection efficiency by introducing a water immersion objective lens to detection optics and employing high-NA light-sheet scanning illumination to confine the illuminated region along the optical axis. As a result, we achieved a signal-to-background ratio approximately 1.8 times higher than that of conventional methods, demonstrating the potential for rapid and high-precision single-cell analysis.
