Solid-state nanopores are expected to be applied to the detection of DNA and other single molecules, but rapid translocation of analytes and clogging events present challenges for measurement. In this study, we analyzed the dynamics of DNA in high-viscosity PEG solutions using fluorescence observation. As a result, we found that in PEG-containing solutions, the entry speed of DNA into the nanopore was reduced, and DNA was in a more condensed state immediately before translocation, compared to standard KCl solution. In addition, the frequency of clogging events was also found to decrease under PEG solution conditions. The influence of PEG solutions on DNA dynamics is discussed.
