Rapid diagnosis of Tuberculosis increases its importance for disease control in some Asian countries. This study aims to detect 85B mRNA of Mycobacterium tuberculosis which is part of the antigen 85 complex. It is essential for the virulence and survival of Mycobacterium tuberculosis. We are developing an early detection method for TB by combining an original high performance isothermal amplification method namely RNA Isothermal Co-assisted and Coupled Amplification reaction and a potentiometricoxideTFT biosensor. In this report,we focus on probe sequence dependance on TFT detection performance, targeting both the outer and inner regions of the RNA amplified product. We designed different types of DNA probe sequences for RNA detection. In this study the outer region targeting probe showed strong signal response between complementary sequence and non complementary sequences.
