A mid-infrared photoacoustic imaging system with a transducer placed on the laser-irradiated side was developed for imaging thick biological samples. Lipid-content mapping was performed on the surface of pork tissue. Spectra acquired from a region containing both lipid and muscle tissues were analyzed by multivariate curve resolution–alternating least squares (MCR-ALS) to extract reference spectra of lipid, muscle, and other components. Multiple regression analysis based on these spectra was used to estimate lipid content. The estimated lipid distribution was in good agreement with stained images, demonstrating the potential of this method for noninvasive visualization of tissue components.
