Peptide self-assembled monolayers on two-dimensional materials serve as molecular scaffolds, yet the correlation between Thioflavin T (ThT) binding properties and amino acid sequences remains unclear. This study evaluated the adsorption characteristics of ThT on h-BN using three peptides with different sequences (VH4, YH4, and LH4) via fluorescence microscopy and AFM to elucidate their interaction mechanisms. Optical and fluorescence imaging of VH4 revealed distinct bright and dark patterns, suggesting the formation of two phases. Comparing the fluorescence intensities of these phases across the sequences demonstrated that LH4 provided the most stable adsorption of ThT molecules. These results clarify the direct impact of amino acid sequences on ThT interactions.
