High-speed atomic force microscopy (HS-AFM) can visualize the dynamics of biomolecules in real time, yet a technique capable of simultaneously measuring the mechanical properties of functioning proteins at high spatiotemporal resolution has not yet been established. In this study, we introduced photothermal excitation using an infrared laser into HS-AFM, developing a method that reconstructs force curves without interference from the instrument's mechanical resonance. By synchronizing the scanning system with the image acquisition system, we achieved simultaneous high-speed topographic imaging and mechanical property mapping.
