In this study, we investigated the effects of horizontal membrane electric fields on the structural and mechanical properties of planar lipid bilayers using bright-field imaging and capacitance measurements. We found that application of a horizontal membrane electric field reversibly decreased both the membrane area and the electrical capacitance, accompanied by a history-dependent response. These findings extend the conventional framework, in which the membrane potential has been treated primarily as a vertical electric field across the membrane, and position horizontal membrane electric fields as a new physical factor governing membrane properties.
