We developed a plasmonic crystal-based sensor using plasmonic crystals fabricated by nanoimprint lithography for the simplefacile detection of the cancer biomarker 5-hydroxymethylcytosine (5-hmC). By utilizing an enzymatic reaction that specifically adds glucose to the hydroxylto hydroxyl group of 5-hmC, we measured the resulting refractive index change via reflection spectrum analysis. Experimental results demonstrated a signal change increase correlated with the 5-hmC content, successfully achieving determinationquantitative detection at a mixing ratio of 10%. This method proves to be useful as a low-cost and high-sensitiveity detection technique.
